  ABSTRACT

CLONING OF VITAMIN D RECEPTOR (VDR) IN pUC19 VECTOR

The vitamin D receptor selectively binds the 1, 25-dihydroxyvitamin D3 [1, 25(OH)2D3] hormone and controls the expression of selected genes in target cells. The vitamin D receptor is a member of the nuclear receptor/steroid hormone receptor superfamily. These receptors function as ligand-activated, transcriptional regulatory proteins. VDR gene was isolated from the blood sample of primary hyperthyroidism patient. The isolated and amplified portion of VDR gene was successfully ligated with pUC19 plasmid DNA to form a recombinant VDR-plasmid construct. That was transformed in to DH5α strain of Escherichia coli. The cloning of fragment of VDR in Escherichia coli was further confirmed by restriction digestion of plasmid isolated from transformed cell culture, by running it on agarose gel electrophoresis.
