  ABSTRACT

FABRICATION OF SOLID LIPID NANOPARTICLES CONTAINING TAMOXIFEN CITRATE AND IMPACT OF PROCESS PARAMETERS ON FORMULATION

CT The objective of present research work was to formulate and characterize Solid Lipid Nanoparticles (SLN) containing Tamoxifen Citrate – Selective Estrogen Receptor Modulator (SERM) and to study the effect of process parameters on Formulation of Solid Lipid Nanoparticles. SLN were prepared by High Shear Homogenization followed by Ultra sonication. Prepared SLN were characterized for Particle Size, Polydispersity Index, Zeta Potential, Entrapment Efficiency, Assay, Morphology and In vitro drug release. Process parameters like homogenization speed, homogenization time and ultra sonication time found to influence the particle size and entrapment efficiency. Prepared SLN were of particle size 95-152 nm and entrapment efficiency 72-85 %. Assay of Tamoxifen in SLN was found to be 99 % in various batches of SLN. SLN were found to spherical in shape by Transmission Electron Microscopy. In vitro drug release study using dialysis membrane indicated SLN as a good carrier for sustained release of drug. SLNs showed sustained release pattern releasing 99.5 % of Tamoxifen over period of 40 hours compared pure drug (240 min). The critical process parameters homogenization speed, homogenization time and ultra sonication time were optimized to get smallest particle size and maximum entrapment efficiency of SLN
