  ABSTRACT

DESIGN AND EVALUATION OF BIOADHESIVE TABLETS OF ROSUVASTATIN CALCIUM

The purpose of this research was to develop bio adhesive gastro retentive drug delivery systems of Rosuvastatin calcium. It is a lipid lowering drug selective and competitive inhibitor of HMG CoA reductase, which reduces the total number of VLDL and LDL particles .which are designed to increase the gastric residence time, thus prolonging the drug release. In the present study, the tablets were prepared by wet granulation technique, using different natural polymers like guar gum, xanthan gum, karaya gum. Thus the study aims to improve the oral bioavailability of the drug and to achieve extended gastric retention which may result in prolonged absorption. The prepared tablets were evaluated with different parameters like hardness, friability, uniformity of weight, content uniformity, swelling characteristics and in-vitro dissolution study. Hardness was found to be in the range of 1.5 to 1.9 kg/cm2, the percent friability was in the range of 0.02% to 0.4% and the tablets showed 40mg of the labeled amount of Rosuvastatin calcium indicating uniformity of content. The tablets containing Rosuvastatin calcium released 96 to 100.8 % of drug at the end of 24th hr by in-vitro release study. The formulation FB2 was selected as an optimized formulation because it gave the best result in terms of the required in-vitro drug release, bio-adhesive study and drug release in extended release manner. Rosuvastatin calcium bio adhesive drug delivery system showed improved bioavailability and extended release which may favor the reduced dose frequency and improved patient compliance
