  ABSTRACT

STUDY THE EFFECT OF PURIFIED URICASE FROM P.AERUGINOSA ON HYPERURICEMIA IN VIVO

Uricase purified from P.aeruginosa was used to study the effect of this enzyme against hyperuricemia. Until now, there are only few studies that concentrate on the effect of uricase enzyme in vivo . The results of the present study showed that regarding hemolysis test ,the purified uricase does not cause hemolysis or erythrocyte agglutination at 37 ºC. Also results of stability of this enzyme in the presence of plasma showed that the enzyme retained about 90-95% of its original activity after 1 and 2 hours of incubation in the presence of plasma, respectively, while only about 40% of the activity was retained after 6 hours of incubation in the presence of plasma. In vivo study revealed that uricase has a potential activity to lower plasma uric acid level from high concentration (298.70 ± 34.91μmol/L) to lower concentration (132.60 ± 14.39μmol/L) within an hour ,consequently this enzyme can be used as a drug protein for hyperuricemia treatment in the future
