EFFECT OF DIFFERENT AUXINS AND CYTOKININS ON IN VITRO MULTIPLICATION AND ROOTING OF SHOOT CULTURES IN SUGARCANE
The effect of different auxins and cytokinins on in vitro multiplication and rooting was investigated in sugarcane (Saccharun spp.) varieties; CoSe 01235 and CoS 99259. To evaluate the response, shoot tip and meristem explants were cultured on Murashige and Skoog (MS) basal medium supplemented with different concentrations and combinations of auxins (NAA and IBA) and cytokinins (BAP and Kinetin). The results showed that MS medium supplemented with BAP and Kinetin (0.5 mg/l each) was best for shoot initiation and culture establishment in shoot tip explant. The best response in terms of multiplication was observed on MS medium with BAP, Kinetin and NAA (0.5mg/l each). Better rooting was recorded, when in vitro shootlets were inoculated on the half strength MS basal media with 5.0 mg/l NAA and 50g/l sucrose. Rooted shoots were transplanted in green house for hardening and their survival rate was 90% in the field condition.
