OPTIMIZATION ON PROTEASE ENZYME PRODUCTION USING Bacillus subtilis SSD-I (JQ747516) ISOLATED FROM LONAR METEORITE CRATER IN INDIA
SSD-I (JQ747516) were isolated from water samples collected from saline Lonar Meteorite crater, (MS) India having pH 10.5. Among them one of the bacterial strains was identified by 16S rRNA sequencing showed 95% alignment match with Bacillus subtilis. The optimum growth of such organism was 12.5 pH and 16% salt concentrations. Chemical analysis of water shows metal ions and salt concentrations in Lonar crater. The Bacillus subtilis have potential to produce proteolytic enzymes such as protease. Among the different sources glucose was found to be best carbon source and Yeast extract was found to be the optimum nitrogen source for protease enzyme production by SSD-I. The optimum enzyme activity was at 10 pH 500C temperature. Soya cake found to be the excellent enzyme substrate for enzyme production among different agro based wastes for test strain grown under submerged fermentation conditions.
