  ABSTRACT

AMELIORATION OF DIABETES INDUCED MEMORY DETERIORATION AND OXIDATIVE STRESS CHANGES WITH TREATMENT OF TRIGONELLINE

Trigonelline is alkaloid obtained from Trigonella Foenum-graecum L. Alkaloids are well known to possess different pharmacological activities and antioxidant properties. Trigonelline known to have different pharmacological activit ies such as antidiabetic, estrogen like, antinocecptive and antineoplastic activities. However, effect of Trigonelline in diabetes induce d memory dysfunction and related biochemical oxidative stress has not been studied. Therefore, study is designed and conducted to evaluate diabetes induced memory loss and oxidative changes in various brain parts of Streptozotocin induce diabetic rats. Diabetes was induced by streptozotocin (60 mg/kg, i.p.). After confirmation of diabetes acute /chronic treatment of Trigonelline (25, 50, 100 mg/kg, p.o.) was started in separate group of animals. The results reveals that after 30 days of diabetes induct ion induced marked memory deficits and also increased cholinesterase and MDA levels and depletes reduced glutathione levels in hippocampus and cerebral cortex. Further, chronic treatment with Trigonellie has significant influence on memory at 50 and 100 mg/kg, p.o doses in diabetic rats. Treatment with Trigonelline significantly reduced blood glucose levels and MDA levels. Trigonelline also elevated GSH levels and significantly reduced evoked cholinesterase levels in diabetic rats. The result obtained from study points that treatment with Trigonelline in diabetic rats ameliorates memory loss and oxidative stress
