MEDIA OPTIMIZATION FOR LIPASE FROM Halomonas salina ku-19, AN EXTREME HALOPHILES ISOLATED FROM LITTLE RANN OF KUTCH
Altogether, thirty extreme halophiles were obtained from salt, mud and water samples collected from little rann of Kutch after enrichment in halophilic broth and complete media broth. On the basis of primary screening on solid media containing tributyrin, Ku-19 was potential lipase producer. 16s r-RNA sequencing data revealed that organism was Halomonas salina. Further optimization studies were performed for lipase secretion in liquid media. Organism was able to secrete maximum lipase after 264 hours of incubation, 15-20% NaCl, pH 7, 40ËšC temperature and 4% substrate concentration.
